Maturations-Inhibitor


Maturations-Inhibitoren (Reifungshemmer) sind Arzneistoffe aus der Gruppe der Virustatika.

Geschichte

An Maturations-Inhibitoren wird schon seit langem geforscht. In der Entwicklung dieser Wirkstoffklasse kam jedoch erst 2005 der entscheidende Durchbruch.

Wirkung

Maturations-Inhibitoren greifen in einem späten Stadium der Virusvermehrung ein und verhindern den Zusammenbau neuer Viruspartikel innerhalb der Zelle, aus den viralen Bausteinen. Ein Virus-Vorläuferprotein wird durch die Virus-Protease in unterschiedliche Funktions- und Strukturproteine aufgespalten. Der Maturations-Inhibitoren binden an das Vorläuferprotein (z.B. CA-SP1) und inhibitiert die Spaltung durch die HIV-Protease. Dadurch kann keine funktionsfähige Virus-Proteinhülle aufgebaut werden.

Nebenwirkungen

Erste klinische Studien an Wirkstoffen dieser Substanzklasse zeigten eine gute Verträglichkeit. Lesen Sie dazu auch die Hinweise bei den einzelnen Wirkstoffen.

Anwendungsgebiete

Maturations-Inhibitoren werden zur Bekämpfung von Viren eingesetzt. Derzeit existieren Wirkstoffe gegen HIV.

HIV Therapie

Maturationsinhibitoren sind, im Rahmen einer HAART, eine interessante neue Substanzklasse. Dennoch müssen sie erst in Phase IIb/Phase III-Studien bestehen. Ob es Wirkstoffe wie Bevirimat in die Klinik schaffen, bleibt unklar. Neue Hoffnung gibt die ersten Studien [1] erwiesene Wirksamkeit. Damit ist klar, dass HIV auch in der Klinik von einer neuen Seite attackiert werden kann.
